Quickest takes into account the road speeds, so a quicker route maybe a motorway route as opposed to an A or B road route.

Shortest is the shortest distance in miles and kilometers.

Shortest will take least miles, quickest will keep you more on major roads, which are generally faster and therefore quicker, although the distance travelled will be further.

One takes you the quickest route (not necessarily the shortest) the other takes you the shortest route (which may not be the quickest)

A
|
B---C

So to get from A to C
Via B may be the quickest as it is all fast roads but it is clearly not the shortest by distance, whereas getting from A directly to C is shortest but it may take longer as the roads are slower.
